Atrial natriuretic peptide deposited as atrial amyloid fibrils

Summary

Deposition of amyloid in the atria is exceedingly common in the aging heart. We have extracted amyloid fibrils from atria and purified a major protein which had N-terminal amino acid sequence identical to that of atrial natriuretic peptide (ANP). Antisera to ANP and to the amyloid fibril protein both labelled atrial muscle cells and atrial amyloid in an identical way.

The amyloid-forming protein ANP is a prominent component in aging-related (senile) atrial amyloidosis, and some studies support the presence of BNP in these deposits as well (20,22,36).       IAA is generated by the local, atrial, deposition of atrial natriuretic peptide (ANP), a substance synthesized by atrial myocytes (Kaye et al., 1986). Johansson et al. (1987) showed that amyloid atrial fibrils contain the same N-terminal amino acid sequence of the ANP, and that antisera to ANP and amyloid fibril protein, label atrial muscle cells and atrial amyloid in an identical fashion. Takemura showed, using immunocytochemistry at the electron microscopic level, that ANP is secreted by atrial myocytes, and confirmed the results using in-situ hybridization for ANP mRNA (Takemura et al., 1998).
